Project Description:
Drugs are administered in a specific dosage and adjusted by physicians to keep an appropriate concentration of the medication in the patients' blood to achieve the desired therapeutic effect without inducing toxicity. However, the dose that would heal and the dose that will harm may be relatively close to each other for certain drugs and even with the same dosage, the therapeutic effect of the medication and its metabolism in the body may differ from patient to patient.

 

Clinicians use therapeutic drug monitoring (TDM) to determine the amount of drug in a patient's blood and use this information to adjust the dosage to ensure safety and treatment efficacy. Nowadays, clinics perform TDM in centralized labs, utilizing sensitive yet slow and expensive analytical techniques, which limits the usage of TDM in resources limited environments and when quick response is needed. 

 

This thesis presents two cost-efficient and fast (less than 15 min analysis time) analytical methods based on surface-enhanced Raman spectroscopy (SERS) for TDM. The first method used a nanostructured SERS sensor for both sample (serum) pretreatment and rapid and sensitive detection, while the second approach combined electrochemistry and SERS to achieve a more sensitive yet still rapid detection. Both methods were also combined with a portable, reasonably priced Raman system, demonstrating the possibility for cost-efficient SERS-based TDM.
