Project Description:
It has become evident that the immune system plays a significant role in cancer development and clearance. Therefore, cancer immunotherapy that involves treatment strategies that provoke the immune system to attack tumor tissue is attractive and anticipated to improve current conventional cancer therapy. This Ph.D. project aims to establish a localized gene therapy platform by creating a transfection system for pro-inflammatory cytokines. Localized gene therapy can be achieved by releasing transfection agents from a biomaterial formulation and using the cellular machinery within the tumor to generate the cytokine in situ. The local production of cytokines will induce a pro-inflammatory reaction in the tumor to help the immune system recognize and eradicate cancer cells.

Perspective:
Today, many different immunotherapeutic approaches have already been developed, with different outcomes in eliminating tumor tissue. For example, immunomodulatory drugs administered systemically have been associated with severe side effects. This local delivery of gene therapy that facilitates prolonged controlled transfection in the tumor might benefit the patient with fewer side effects and improve the patient response rates to anti-cancer immunotherapy
