Project Description:
The main objective of this project is to develop a 3D printed stem cell differentiation and tissue engineering platform for mimicking human cardiac muscle in vitro. The miniature muscle tissues cultured on the platform will be used for evaluating cardiotoxic effects of compounds with potential non‐acute cardiotoxic mechanisms.
 
Perspective:
Organs‐on‐chips systems aim to replicate physiologically relevant human tissues allowing toxicity and efficacy tests on human derived tissue well before clinical trials. Yet, current systems have numerous shortcomings such as consistent fabrication and long-term data acquisition. Multi‐material 3D printing is a powerful tool for addressing these challenges. It enables fabrication of tailored soft, electronic platforms for culturing miniature cardiac tissues during a prolonged period as well as for monitoring tissue contractility in a non-invasive manner. This will support identification of compounds with non‐acute yet fatal cardiotoxicity in long-term exposure, such as chemotherapeutic agent doxorubicin. Furthermore, using patient-derived pluripotent stem cells on this platform patient-specific drug response can be monitored.
