Project Description:

Dendritic cells (DCs) are the major antigen presenting cells of the immune system and play a pivotal role in immune activation versus tolerance in the small intestine. DCs are classically divided into two major subsets: cDC1 (CD11b-CD103+) and cDC2 (CD11b+CD103+) DCs. Transcription factors (TFs) network direct lineage fate of all immune cells. We have previously shown that deficiency in the transcriptional repressor Bcl6 leads to the absence of CD11b- cDC1 DCs across all organs. We here characterize the DC populations remaining in mice lacking Bcl6 in all DCs (DC-Bcl6KO) or in cDC1 DCs (cDC1-Bcl6KO) specifically.

Perspective:
We are currently performing RNA-Seq, ATAC-Seq and functional studies to investigate the direct and indirect genetic targets on Bcl6 and the consequences of Bcl6 deficiency on DC subset functionality. Our work will hopefully contribute to the understanding of the complexity of TF networks driving DC subset biology.
