Project Title: The Role Of The Transcription Factor T-Bet In B Cell Differentiation And Function
Group: Mucosal Adaptive Immunology
Supervisor: Bengt Johansson Lindbom

Project Description:
The main object of this project is to understand the function and phenotype of B cells expressing the transcription factor T-bet. These cells have been related to some autoimmune diseases before and ongoing studies show that they have important roles in some other conditions. In this project, we will try to characterize them both in mice and human intestinal surgical material and lymph nodes from healthy individuals and IBD patients. Since they were discovered relatively recently, our understanding of these cells are limited. We will also try to understand the signaling pathways related to these cells and their interactions with helper T cells.

B cells contribute to immune response by producing antibodies but it is also known that inappropriate B cell responses are associated with wide range of immune mediated inflammatory diseases. T-bet expressing B cells are associated with a number of auto-immune or inflammatory disorders, including systemic lupus erythematosus (SLE) and Crohn’s disease (CD). We will try to understand these cells using multi-color flow cytometry, single cell RNA sequencing and mice models with conditional knockouts thereby contributing to understanding and possible treatments of conditions related to these cells.
